چکیده

Protein permeability of the ciliary epithelium of enucleated arterially perfused cat eyes was studied. Tracer concentrations of 125I-albumin, in an arterial perfusate solution consisting of a balanced salt solution (BSS) without added colloid, was found in the anterior chamber fluid (AH) in a percentage similar to the AH/plasma ratio found for protein in the living cat eye. Stimulation of AH formation by acetylcholine plus eserine increased protein permeability markedly and by a similar amount in eyes perfused either with BSS or with 100% serum. These data indicate that arterial perfusion of the enucleated eye with BSS alone induces no gross abnormality of the blood-aqueous barrier.

Absolute sensitivity of the electroretinogram and of the optic nerve action potential in the perfused feline eye.

PURPOSE To assess and characterize the electroretinogram (ERG) and the optic nerve response (ONR) at threshold stimulus intensity in the isolated perfused cat eye. METHODS Eyes were enucleated from deeply anesthetized adult cats and arterially perfused with oxygenated serum-enriched tissue culture medium. Light stimuli of 20- to 400- msec duration from a xenon arc source, attenuated down to t...

Ocular ischemia and the effects of allopurinol on functional recovery in the retina of the arterially perfused cat eye.

PURPOSE This study sought to examine the acute effects of ocular ischemia and reperfusion on retinal function and determine the extent to which recovery during reperfusion is improved by allopurinol (AP), a blocker of xanthine oxidase (XO). The latter is presumed to be a major factor in the formation of free radicals associated with reperfusion of ischemic tissue. METHODS Electroretinographic...

Theophylline abolishes the light peak in perfused cat eyes.

Theophylline, a phosphodiesterase inhibitor, was applied to the arterially perfused cat eye in vitro. At a concentration of 850 microM, theophylline abolished the light peak of the DC-ERG and led to a 20% increase in perfusate flow to the eye. The effects were reversible. An experimentally-induced increase in flow per se by 20% resulted in a slight enhancement of the light peak. The data sugges...

Effect of prostaglandins on the blood-aqueous barrier of the perfused cat eye.

With a technique of direct visualization of the arterially perfused cat eye, the pressure head may be chosen so that fluorescein added to the perfusate just barely stains the ciliary processes. After addition of PGE1, PGE2, PGF2alpha arachidonic acid, or indomethacin, with or without PG's, no more dye emerged from the processes. The addition of acetylcholine in the eserinized eye floods the pro...

Adenosine: autoradiographic localization and electrophysiologic effects in the cat retina.

Autoradiography with 3H-adenosine was used to localize cells that accumulate adenosine in the cat retina. Electrophysiologic effects elicited by adenosine on DC-electroretinograms (ERG) and optic nerve responses (ONR) were studied in isolated, arterially perfused cat eyes. Subpopulations of cells localized in the ganglion cell layer and inner nuclear layer showed clear labeling for adenosine. T...
